Introduction: Peripheral nerve damage is a common problem in the clinic. These damages can cause severe disabilities and decrease quality of life and they may be costly. Nowadays Melatonin and Curcumin are widely used for treatment of nerve injuries because of their antiinflammatory, antioxidant and neuroprotective properties. The aim of the current study was to compare the effects of Melatonin and Curcumin on the repair of sciatic nerve crush injury.
Materials and Methods: In this study, 40 adult male rats were randomly divided into 5 groups (n=8 per group), including Sham (normal rats), Injury (The rats whose sciatic nerve were crushed for two minutes with a forceps), vehicle (injury + intraperitoneal injections of DMSO(5%V/V) for 4 weeks) and treatment groups (injury + intraperitoneal injections of 100 mg/kg Curcumin and 10 mg/kg Melatonin for 4 weeks). functional test (SFI) was performed during the study period and on days 1, 7, 14, 21 and 28 to evaluate sciatic nerve function. Finally, after 4 weeks, the rate of nerve repair was evaluated using the following techniques: Measurement of wet weight of gastrocnemius muscle in each rat, Hematoxylin-Eosin (H&E) staining to study the diameter of gastrocnemius muscle fibers, Luxol fast blue (LFB) staining for examination of myelin in sciatic nerve axons and Immunohistochemistry technique with anti-Schwann antibody (S100) to evaluate Schwann cell number in sciatic nerve.
Results: our data analysis showed that curcumin and melatonin treated groups had better results than vehicle and injury groups and their difference was significant(p<0/05). Among the treatment groups, the curcumin group showed better results than the melatonin group and was similar to the sham group.
Conclusion: according to this study, Melatonin and Curcumin can be used to accelerate neural recovery of nerve injury. In addition, Curcumin had better function than Melatonin on induction of nerve repair.
